3D Structured Illumination Leads to Resolution Doubling in Widefield Microscopy. M. Gustaffson and colleagues have extended their structured illumination method into the third dimension to also increase the axial resolution of widefield microscopes by a factor of two. Three mutually coherent light beams are generated by a grating and form a three-dimensional illumination and interference pattern. The spatially structured excitation causes high-resolution information to be encoded in the detected images.
