Raman spectroscopy uses the energy shift of a laser-beam that occurs when it is inelastically scattered by the molecules in a sample. As the shift is characteristic for each molecule, the components in the sample can be easily identified. If this fast and non- destructive technique is combined with imaging techniques such as light-, fluorescence-, confocal-, electron-, or atomic force-microscopy, it enables the identification and localization of all chemical components of a sample in a single experiment.

A complete Raman spectrum is acquired at each pixel in the image, and then integrated to generate false color images based on the material’s composition and structure. Raman imaging is widely used to characterize the distribution of components within a sample, but it is also useful for investigating material concentration, phase, stress/strain, and crystallinity.

Together with the Raman experts at WITec, the team at Imaging & Microscopy produced an infographic showing the basics and applications of correlative Raman imaging.

Using groundbreaking research [1] on the visualization of biochemical markers of atherosclerotic plaque in cross sections of brachiocephalic arteries (BCA) as an example, the infographic additionally shows the capabilities of Raman imaging in the Life Sciences.
