Live-cell Super-resolution Imaging
R. Wombacher and co-workers have generated a chemical tag (trimethoprim, TMP tag) that can be used for direct stochastic optical reconstruction microscopy (dSTORM). A monomeric TMP binding protein that is significantly smaller than fluorescent proteins can be fused to the protein of interest and will bind the cell-permeant TMP-ATTO665 fluorophore conjugate that can subsequently be imaged. The potential of the approach is demonstrated by visualizing human histone H2B in vivo at a resolution of approx. 20 nm.
